Research experience: Main areas of research have been (in chronological order): Trombosis and haemostasis, including coagulation and monoclonal antibodies against receptors on blood platelets and coagulation factors. Experimental cancer research in murine model systems, involving anti-idiotypic antibodies for therapy of B-cell tumours, and the role of NK-cells and macrophages in tumour immunology. Production of monoclonal antibodies against human cancer cells and cells of the immune system. Production of monoclonal antibodies against HLA molecules for the study of the role of these molecules in peptide specificity, T cell recognition, tumour immunology and transplantation. Mechanisms of host-tumour recognition; Studies on the function of HLA molecules, T cell subsets and antigen presenting accessory cells in tumour immunology and transplant rejection processes. Production of monoclonal antibodies against haematopoetic progenitor cells for leukaemia phenotyping, studies of early haematopoiesis and as tools in gene therapy directed at haematopoetic progenitor cells. Development of new methods for isolation of pure cell subsets including haemopoietic stem cells by magnetic immunobead technology. Clinical cancer immunology, including studies of the immune system in various groups of cancer patients. Specific immunotherapy; Development of cancer vaccines targeting mutant oncogenes, including RAS, TGFβRII and other oncogenes/tumor suppressor genes, and characterization of the HLA molecules and T cells involved in oncopeptide responses. Translational research: Clinical trials; testing of vaccine candidates in phase I/phase II clinical trials. Gene therapy/immuno-gene therapy, including cancer vaccines based on engineered dendritic cells transfected with tumour mRNA and reprogrammed using cytokine mRNA and siRNA for gene silencing and re-directed T cells engineered by transfection of mRNA encoding chimeric receptors and selected T cell receptors. Immunogenomics: High througput technologies for tumor antigen discovery, including SEREX and fully automated SEREX with filamentous phage tumor cDNA expression libraries. Carcinogenesis and molecular pathology, including genetics and epigenetics of colorectal cancer. Immunogenetics/pharmacogenetics: Role of SNP's and other polymorphisms in determining immune responses to cancer vaccines and in tumor development. Development of immunotherapy targeting cancer stem cells. Targeting of nanoparticles to micrometastasis and cancer stem cells.
Scientific highlights: Generation of a large panel of reference monoclonal antibodies against haematopoietic stem cells (1987). First detection of spontaneous immune responses in cancer patients against point mutation in ras oncogene (Int. Immunol. 1992). First publication of a clinical trial (first in man) of a peptide vaccine in cancer patients (The Lancet 1995). Comprehensive model of Carcinogenesis and natural selection (Adv. Cancer Res. 1999). First detection of spontaneous responses against a new class of frameshift mutations in colon cancer patients (PNAS 2001), opening for prophylactic vaccines for patients with genetical risk of colorectal cancer. Workshop participation: Submitted a number of own monoclonal antibodies to and actively participated in several of the International Workshops on Human Leucocyte Differentiation Antigens and the International Histocompatibility Workshops. Several of these antibodies are now reference CD antibodies or HLA reagents.
Clinical trials: Organization of and participation in more than twenty-five investigator initiated clinical trials of tumor vaccines and immuno-gene therapy. These trials include the first study of a ras peptide vaccine (1993) and the first study of telomerase peptide vaccination in cancer patients (2000), seven immuno-gene therapy trials with mRNA transfected dendritic cells.
Scientific cooperation: Member of CEVI (center for vaccine development and immunotherapy, Medical Faculty, University of Oslo). Recent project cooperation with Affitech AS on the identification on pancreas, prostate and breast cancer antigens by the use of high throughput phage technology.
